類胡蘿蔔素在色素市場上佔據了很重要的地位,不管是添加於飼料、著色劑還是在醫療保健。而在類胡蘿蔔素中有種紅色色素-蝦紅素,因具備強力的抗氧化能力,近年來常被拿來進行疾病與癌症的相關研究。先前於南台灣分離出一株微藻,命名為Coelastrella sp. F50,當遭遇到逆境時藻體會累積橘紅色色素,經由高效液相層析儀(high performance liquid chromatography)分析得知那些色素中含有蝦紅素、角黃素、葉黃素以及β-胡蘿蔔素,後續實驗探討如何以實驗室條件達到蝦紅素最大生產值。在皂化實驗中顯示,10 mM的氫氧化鈉為最適合的濃度,經過兩小時皂化後會有最大值的游離蝦紅素。在光強800 μmol/m2s下能有效刺激藻細胞累積色素,而在培養過程中添加碳酸氫鈉(2 g/L),色素累積成效會優於二氧化碳曝氣的培養組。經過14天養殖,重量可從100 mg/L增加至2 g/L,蝦紅素最高含量可達2.1%,每公升約可生產42 mg/L蝦紅素,生產率更可達到3 mg/L/day,從數據來看,Coelastrella sp. F50不但具耐熱性且具備高產率的特質,或許能替代雨生紅球藻作為熱帶地區生產蝦紅素的藻種。
Abstract
Carotenoids occupy an important niche in current natural pigments market. They can be used as feed additive, food colorants, cosmeceutical or dietary supplement.. The application and effect of astaxanthin are actively studied due to its high antioxidant capacity in recent years. A new species of thermotolerant microalga was isolated in tropical Taiwan, and named as Coelastrella sp. F50. These cells produce reddish pigments under environmental stress. The pigments contain astaxantin, canthaxanthin, lutein and beta-carotene analyzed using high performance liquid chromatography. Productivity of astaxanthin in this species was investigated. Optimization of saponification conditions demonstarate that 10 mM NaOH mM was the optimal concentration to react for two hours. Optimization of irradiance stress shows that 800 μmol/m2s leads to the highest astaxanthin accumulation in the cultures. Bicarbonate (NaHCO3) enhanced astaxanthin accumulation, and the level of 2 g/L had the best effect. In 14 days of cultivation, biomass of the cells increase from initial 100 mg/L to 2 g/L and maximum astaxanthin content reached 42 mg/L (2.1%). The maximum astaxanthin productivity reached 3 to 15 mg/L/day depending on different cultivation scenarios. Based on the results, Coelastrella sp. F50 has a chance to commercially produce natural astaxanthin in tropical areas. More studies are needed to further optimize astaxanthin production using this new microalga.
