電噴灑輔助雷射脫附游離:法 ( Electrospray-Assisted Laser Desorption Ionization, ELDI ) 是一種在大氣壓力下進行的質譜游離方式。在進行電噴灑輔助雷射脫附游離法分析時,會利用高能聚焦脈衝雷射將樣品溶液脫附成微小液滴,藉由電噴灑溶液所形成的帶電液滴與分析物融合後使分析物帶電荷進入質譜儀分析。

  對於直接液體樣品的分析,先前研究是利用能量小的UV雷射進行雷射脫附(~100 μJ),但雷射會穿透液體的表面,導致能量過小使得樣品難以脫附。因此會在樣品中添加碳粉後再以雷射轟擊液滴表面,藉由溶液中碳粉吸收雷射光能量而使得分析脫附。而分析樣品中的碳粉會與分析物一同進入質譜儀造成質譜儀內部汙染。

  本論文研究中發現,改為能量大的雷射(1 mJ)進行脫附,則能夠迅速的將樣品脫附,但分析物的訊號持續的時間十分短暫(<30s)。為了增加分析物訊號能夠持續的時間長短,研究第一部分主要探討將分析樣品滴於在各式種類的基材上進行雷射脫附。並且將樣品於固定位置上偵測訊號持續時間,以找出可以延長樣品的持續時間且不須添加碳粉的方式進行脫附。
  第二部分針對電噴灑輔助雷射脫附游離法應用於不織布樣品載具下之參數探討,其中包括雷射能量強度、雷射頻率、不同樣品含量、液體黏滯度與不同溶劑組成的探討等,以找尋在不織布樣品載具下最佳化之結果,最後應用各種最佳化參數探討其偵測極限,最低可以偵測約10-7M。

  第三部分乃將不織布樣品載具應用於各式不同的樣品分析物上,由於針對分析樣品的持續延長,因此可以有更加充裕的時間進行分析樣品中不同m/z 樣品之探討。首先利用五種不同顏色染劑標準品所混合而成樣品分析物在固定位置上持續的利用雷射脫附所得到的訊號在持續進行的時間下個別進行MS/MS 分析,可以成功的區別出五種不同樣品之結果。接下來將樣品應用到各式生化樣品下,皆可以得到長時間的持續時間,因此也能夠將不織布樣品載具應用生化樣品溶液上。最後將不織布樣品載具應用到水果汁液分析,分別利用正離子與負離子偵測,在持續的樣品訊號時間下,偵測果汁的成分結果。結論利用了不織布樣品載具做為樣品平台可以使得訊號的持續時間增長,能夠有更充裕的時間對分析樣品僅行分析,且所需要的樣品含量少(~15 μL),能夠減少樣品的消耗,並可以於短時間下快速的進行分析。
