本研究主要是從台灣小琉球採集的軟珊瑚 Sinularia flexibilis的有機萃取物中尋找具有生物活性的二次代謝物。從這株珊瑚共分離得到十五個天然化合物1–15,包括五個新的cembranolide-related化合物Flexibilisin D (1)、Secoflexibilisolides A and B (2 and 3)、Flexibilisolides H and I (4 and 5),十個已知化合物 616。所有化合物的化學構造及相對立體組態均由光譜數據的分析 (IR、MS、1D、2D NMR) 和比對文獻上已知化合物的光譜資料而決定。
本研究中,將所獲得的化合物1, 3–16進行小鼠血癌細胞 (P-388)、人類慢性白血病细胞 (K-562)、人類結腸癌細胞 (HT-29) 的細胞毒殺活性測試與抗發炎活性測試。其中,化合物7, 10, 11對小鼠血癌細胞具有中等的細胞毒殺活性,在化合物濃度為10 μM時,IC50抑制濃度分別為24.1, 16.2, 16.0 μM;而抗發炎實驗的部分,只有化合物11有抑制效果。
Abstract
In order to discover bioactive secondary metabolites, we have studied the chemical constituents from the organic extracts of soft coral Sinularia flexibilis. This study had led to the isolation of fifteen natural compounds 1–15, including five new cembranolide-related compounds, Flexibilisin D (1), secoflexibilisolides A and B (2 and 3), flexibilisolides H and I (4 and 5) along with ten know compounds from S. flexibilis. The structures and relative configurations of all metabolites had been established by detailed spectroscopic analysis including 1D and 2D NMR (1H, 13C, 1H–1H COSY, HSQC, HMBC and NOESY) data in association with MS experiments.
The cytotoxicity of compounds 1, 3–16 against the P-388 (murine leukemia), K-562 (human erythro myeloblastoid leukemia) and HT-29 (human colon carcinoma cells) were assayed, and compounds 7, 10, 11 showed significant cytotoxicity against the P-388 cell line with IC50 values of 24.1, 16.2 and 16.0 μM, respectively. On the part anti-inflammation assay, compound 11 were found to show inhibiting activity.
