中文摘要
由於蛋白質的基本構成為氨基酸,因此氨基酸的性質,如親水、疏水、帶電、酸鹼性、鏈長...等性質都會對水合位置和數目造成重要的影響,因而我們利用固態核磁共振實驗方法以及鬆弛速率的測量,從定量上來了解這些特性如何影響水合行為。我們製備一系列不同配比的水合氨基酸樣品,來觀察其在光譜以及動態的變化,並希望由本實驗所建立起的資訊可以提供蛋白質水合研究上做為一個定性與定量的參考。
本研究的重點在於改變不同的水合程度的氨基酸,測得其縱向及橫向的鬆弛速率,並結合二維光譜的方法以及其他固態實驗方法來進一步確認結合水的數目及位置。最後藉由這些定量的數據建構出一個水合氨基酸分子的模型,以利未來理解類似條件下的蛋白質水合特性。
