七個新eunicellin-based 雙萜化合物 krempfielins K-Q (1-7),伴隨著六個已知化合物,litophynin I (8),krempfielin A (9),krempfielin D (10),sclerophytin F (11),litophynol B (12) 和 litophynol I monoacetate (13),是從軟珊瑚 Cladiella krempfi中分離出來,這株軟珊瑚是從澎湖採集而來的。這些化合物的結構是由光譜數據分析 (IR, MS, 1H NMR, 13C NMR, HSQC, HMBC, COSY, and NOESY) 並比對文獻上已知化合物之光譜數據而定的。化合物1的結構較少見,是由於從C-2與 C-6 經氧原子連接組成 tetrahydropyran ring。化合物7是目前第一次發現為 eunicellin-based 的 dimer。
化合物 1–6 和 8–13 以 10 μM 的濃度進行抗發炎活性測試。結果顯示,化合物 4 可以抑制超過 70% 在 FMLP/CB-induced human neutrophils 的 elastase 釋放表現。
Abstract
Seven new eunicellin-based diterpenoids, krempfielins K-Q (1-7), along with six known compounds, litophynin I (8), krempfielin A (9), krempfielin D (10), sclerophytin F (11), litophynol B (12), and litophynol I monoacetate (13), have been isolated from the soft coral Cladiella krempfi which was collected from Penghu Island of Taiwan. Their structures were determined through extensive spectroscopic data (IR, MS, 1H NMR, 13C NMR, HSQC, HMBC, COSY, and NOESY) and by comparison of the spectral data with those of the related known compounds. The structure of compound 1 is rare due to the presence of a tetrahydropyran ring constituted from C2-C6 and oxygen atom. Compound 7 is the first discovery of eunicellin-based dimer.
The anti-inflammation activity of twelve compounds (1–6 and 8–13) at 10 μM was studied. The results showed that compound 4 inhibited more than 70% express of elastase released in FMLP/CB-induced human neutrophils .
