其中從傘軟珊瑚Cespitularia hypotentaculata 分離純化得5個新的diterpene 化合物,分別為GN67-12-5-4-2 (1)、GN67-12-6-4-2 (2)、GN67-20-4-3 (3)、GN67-25-6-1 (4)、GN67-12-5-12-1 (5) 及4個已知的化合物分別為 GN67-12-5-12 (6)、GN67-28-5-3-3 (7)、GN67-28-5-4 (8)、GN67-23-4 (9),其中GN67-25-6-1 (4) 可以形成結晶,具有 X-ray 結晶繞射輔助化學位向的佐證,化合物 5 經由做 Mosher 酯化反應決定其絕對立體化學。
從仿穗軟珊瑚 Paralemnalia thyrsodes 中分離純化得3個新的sesquiterpene化合物,分別為 GN78-14-3a-B (10)、GN78-22-8I-B (11)、GN78-17-3a-B’ (12) 及5個已知的化合物分別為GN78-12-5-B-G (13)、GN78-14-3a-N (14)、GN78-22-8-W (15)、GN78-22-8-H (16)、GN78-24-5-Q (17)。
另外也從多刺棘穗軟珊瑚 Dendronephthya divaricata 中分離純化得3個已知的化合物分別為 GN72-9-8-J (18)、GN72-7-4-2 (19)、GN72-19-6-8-E (20)。
以上這些化合物的結構皆是經由大量廣泛的光譜分析及化學反應而建立。在進行抗發炎活性測試當中,其中化合物 17 對發炎性蛋白 iNOS (誘發型一氧化氮合成酶) 具有抗發炎活性。
Abstract
Five new diterpenes, GN67-12-5-4-2 (1)、GN67-12-6-4-2 (2)、GN67-20-4-3 (3)、GN67-25-6-1 (4)、GN67-12-5-12-1 (5) and four known terpenoides, GN67-12-5-12 (6)、GN67-28-5-3-3 (7)、GN67-28-5-4 (8)、GN67-23-4 (9) were isolated from the methylene chloride solubles of the Formosan soft coral Cespitularia hypotentaculata.
Three new cytotoxic sesquiterpenes, GN78-14-3a-B (10)、GN78-22-8I-B (11)、GN78-17-3a-B (12) and five known terpenoides, GN78-12-5-B-G (13)、GN78-14-3a-N (14)、GN78-22-8-W (15)、GN78-22-8-H (16)、GN78-24-5-Q (17) were isolated from the Formosan soft coral Paralemnalia thyrsodes
Three known compounds, GN72-9-8-J (18)、GN72-7-4-2 (19)、GN72-19-6-8-E (20) were isolated from the methylene chloride solubles of the Formosan soft coral Dendronephthya divaricata
The structure of 4 was further confirmed by X-ray crystallographic analysis and the configuration of the hydroxyl at C-6 of 5 was further determined by Mosher’s reactions. It was suggested that the C-6 has the S configuration.
The inhibition of these metabolites toward the pro-inflammatory proteins (iNOS and COX-2) expression was also investigated. Compound 17 showed more specific inhibition against the up-regulation of the pro-inflammatory iNOS protein of LPS-stimulated RAW264.7 macrophage cells.
